Geron Corporation Reports Publication of Research Supporting the Utility of Telomerase as a Universal Antigen for Cancer Immunotherapy  


MENLO PARK, Calif., Mar 18, 2003 (BUSINESS WIRE) -- Geron Corporation
(Nasdaq: GERN) announced today the publication of research results that support
the utility of telomerase for widespread applications in cancer immunotherapy.

The research, published in the March 2003 issue of Cancer Gene Therapy, shows
that different types of cancer cells, including cancer cells without any known
tumor-associated antigens, can be killed by telomerase-based immunotherapy.
Geron scientists studied melanoma and colon cancer cell lines that are known to
express the tumor-associated antigens, MART-1 and CEA-1, respectively, and an
osteosarcoma line lacking any known tumor antigen. Telomerase was introduced
into human dendritic cells by gene transfer enabling the dendritic cells to
generate telomerase specific cytotoxic T-lymphocytes (CTLs) that killed all
three tumor cell lines. These findings show that telomerase vaccination can be
used in all cancer patients, even those with tumors that do not express any
known tumor-associated antigens.

Telomerase is activated in all human cancer types, including breast, lung,
colon, prostate, pancreatic and hematologic tumors. Telomerase is therefore an
attractive candidate for use as a universal therapeutic cancer vaccine. A Phase
1 study of Geron's ex-vivo telomerase vaccine is currently underway in patients
with metastatic prostate cancer at Duke University Medical Center.

Study Results

In the study, Geron scientists were able to show that dendritic cells (the most
potent antigen presenting cells in the body) obtained from the peripheral blood
of normal healthy volunteers can be modified in vitro with purified human
telomerase DNA or with an adenoviral vector containing human telomerase DNA. The
resulting telomerase-modified dendritic cells then present antigenic fragments
of the telomerase protein to T-lymphocytes, thereby enabling them to recognize
and kill telomerase-positive cancer cells. This study shows that dendritic cells
genetically modified with the telomerase gene can be used to generate an immune
response against cancer cells, and extends previous work that showed similar
results using telomerase RNA (Nature Medicine 2000;6:1011-1017) or telomerase
peptides (Proc. Natl. Acad. Sci. USA 2000; 25:4796-4801) as antigens. These
results support the potential use of telomerase as a universal cancer vaccine
administered directly to cancer patients in order to induce a strong and
specific anti-telomerase immune response in which the patients own lymphocytes
recognize and kill telomerase-expressing cancer cells.

viruses (partnered with Genetic Therapy, Inc./ Novartis), and telomerase

Geron Shares Rise as Firm Gains Footing in Patent Battle

NEW YORK -- Geron Corp. (NasdaqNM:GERN - News) shares rose 15% Monday on hopes that the company has a good chance of winning at least one part of its three-way patent battle over the intellectual property surrounding animal cloning.

Geron has been locked in a three-way battle with Advanced Cell Technology, based in Worcester, Mass., and Infigen Inc., DeForest, Wis., over the rights to the technology that created Dolly the cloned sheep.

Last week, a judge ruled in favor of Advanced Cell in its fight with Infigen.

The judge hasn't ruled in Geron's lawsuits against Advanced Cell or Infigen. But Infigen is using the same claims it used against Advanced Cell in its suit against Geron, so Geron's vice president for intellectual property, David Earp, says last week's ruling has a positive implication for Geron.

Geron isn't involved in animal cloning itself - it is a development-stage company specializing in oncology - but has licensed its intellectual property surrounding animal cloning to other companies. Geron acquired the patents from the Roslin Institute, the Scottish research institute that created Dolly.

Advanced Cell and Infigen officials weren't immediately available for comment.

In afternoon trading on the Nasdaq Stock Market (News - Websites), shares of Geron were up 53 cents, or 16%, at $3.92.

Neuer Ansatz zur Therapie von Krebs
Wissenschaftler wollen Tumorzellen durch das Abfangen des Enzyms Telomerase sterblich machen

Berkeley - Einen neuen Weg bei der Behandlung von Krebs haben möglicherweise Wissenschaftler der University of California in Berkeley geebnet. In der aktuellen Ausgabe des Fachmagazins "Nature Cell Biology" sind die Ergebnisse ihrer Studie veröffentlicht.

Die Forscher haben herausgefunden, dass das für die Unsterblichkeit der Krebszellen verantwortliche Enzym Telomerase in diesen Zellen über den gesamten Zellkern verteilt in aktiver Form vorliegt. In gesunden Zellen ist es dagegen ausschließlich in einem bestimmten Bereich des Zellkerns lokalisiert und wird nur während des Stadiums der Zellteilung freigesetzt. Der neue Ansatz für eine Therapie könnte darin liegen, das Enzym in den Krebszellen wieder einzufangen oder zu hemmen, um ein unbegrenztes Wachstum der Krebszellen zu verhindern.

Das Enzym Telomerase kommt nicht in allen Zellen des Körpers vor, sondern nur in sich besonders schnell teilenden Zellen. Es findet sich normalerweise nur in Keimzellen, den sich ständig erneuernden Zellen der obersten Hautschicht, in Knochenmarkszellen sowie in Gewebezellen, die den Verdauungstrakt auskleiden. Die Telomerase hilft dabei, DNA-Schäden während der Zellteilung zu vermeiden und somit die Aufrechterhaltung der Erbinformation in der nächsten Zellgeneration zu gewährleisten. In 80 Prozent aller menschlichen Tumorzellen ist es vorhanden und überdies in einer besonders hohen Konzentration.

Die Erbinformation des Menschen befindet sich in jeder Zelle des Körpers im Zellkern auf insgesamt 23 Chromosomenpaaren. Die Chromosomen haben für die DNA sowohl Schutz- als auch Trägerfunktionen. Sie haben an ihren Enden kurze DNA-Schutzkappen, ähnlich wie die Plastikhülsen an einem Schnürsenkel. Die so genannten Telomere enthalten keine Erbinformation und bewahren die Chromosomen vor einem Verlust von essenzieller DNA. Mit jeder Zellteilung geht aber ein kleines Stück dieser Schutzkappe verloren, so dass die Telomere kürzer werden. Normalerweise kann sich eine Zelle zwischen 40 und 60 Mal teilen. Dann sind die Telomere auf eine kritische Länge geschrumpft, und die Zelle hört auf, sich zu vermehren, altert und stirbt.

In den Zelltypen allerdings, die Telomerase besitzen, also auch in Krebszellen, werden diese verloren gegangenen Telomerstücke wieder von dem Enzym Telomerase erneuert. Auf diese Weise erhöht sich die Lebenszeit der Zellen enorm. Das Zählwerk wird gewissermaßen abgeschaltet, und die Zelle wird unsterblich.

Die Telomerase ist auch bereits als so genanntes "Unsterblichkeitsenzym" bekannt und gab Hoffnung auf ein mögliches Zurückdrehen der biologischen Uhr. Mit Hilfe von Enzym-Injektionen sollte der Zellalterung Einhalt geboten werden und zu einem längeren Leben führen. Im Reagenzglas konnte die Lebensspanne normaler menschlicher Zellen mit Telomerase bereits verlängert werden.

Weiterhin könnte Telomerase bei der Behandlung von Anämien oder HIV eingesetzt werden, um die Stammzellen im Blut oder des Immunsystems zu stärken. Dennoch müssten nach Ansicht der Forscher noch Wege gefunden werden, wie das Enzym an die richtigen Stellen in der Zelle gelangt, um dort wirken zu können. Allerdings könnten die Stammzellen auch selbst in der Lage sein, das Enzym an den richtigen Funktionsort zu transportieren.

2. Genauso könnte die Telomerase in Krebszellen zerstört/inaktiviert werden, um die Sterblichkeit
dieser Zellen zu erhöhen.
3. Auch auf dem zukunftsweisenden Gebiet der Benutzung von Stammzellen für
Gewebstransplantationen, Gentherapien, etc. sind diese Kenntnisse nützlich.

Endlich wirds mal gehört! Habe bereits bei 1,9 Euro zum Kauf empfohlen, aber keine Resonanz hier bei ariva. Der Wert wird doch sogar vom Staat gefördert, Bush läßt grüßen. Mein Kursziel 12-18 Euro normal. Aber ein wenig Geduld bitte. In der Boomzeit hatten sie schon von 200 Euro langfristig geschnackt, aber das würde ich nun nicht  gleich behaupten. An schlechten Börsentagen legt Geron dauernd zu, weil sich die Jungs bei Kursrückgang sofort eindecken. Noch halte ich den Wert als Schnäppchen, aber bald würde ich persönlich nicht mehr nachkaufen. Stemcells ebenfalls interessant?!

Der Ausbruch vor drei Wochen (über 100% an einem Tag) hat sich damit wohl nicht als Fehlsignal erwiesen. Das sieht jetzt wirklich gut aus - wie auch in der US-Biotech-Branche allgemein, die sich seit einigen Monaten gut hält.
Insgesamt macht der US-Aktienmarkt einen viel stabileren Eindruck als der deutsche. Ich glaube, dass viele Small- und Mid-Caps in den USA mittlerweile ihre Tiefststände gesehen haben.

ne bin nur im Wertpapiergeschäft und Onlinegeschäft tätig. Geron wird noch weiter steigen, wenn keine bösen Überraschungen kommen. Desweiteren ist Viropharma spannend, da sie ein Medikament noch immer in Phase 3 haben sollen. Damals haben sie behauptet, mindestens 1 Jahr bräuchten sie noch. Rate mal wie lange das her ist? 1 Jahr und ein paar Wochen?! Auf jeden Fall wird der Kurs gut gehalten. An einigen Tagen wird sich da heftigts eingedeckt und dann wieder auf 1,45 Euro etwa fallen gelassen. Habe selber nur kleines Paket zur Zeit, bin schon voll investiert.

in Phase 3 soll glaube ich irgend ein  nicht weltbewegendes Erkältungsmedikament sein, deswegen sind die Auflagen angeblich so streng gewesen, daß weitere Tests abverlangt worden. Aber da kennst Du dich wohl wahrscheinlich besser aus. Auf jedem fall startet der Wert jetzt schon durch, aufgrund der Hoffnung?!

Revenues for the quarter totaled $784,000, compared with $1,159,000 for the comparable period in 2002, and $809,000 for the previous quarter ended December 31, 2002. The net loss attributable to common stockholders for the quarter was $6,374,000, or $0.04 per common share, compared with a net loss attributable to common stockholders of $678,000, or $0.02 per common share, for the three months ended March 31, 2002. The net loss for the first quarter of 2003 included $3,587,000 in non-cash charges that were related to the grants of common stock and options and warrants as compensation for services and non-cash interest expense related primarily to the accounting for Calypte's convertible debt financing instruments. During the quarter ended March 31, 2002, non-cash charges and credits were a net credit of $1,292,000, related primarily to the gain on settlement of debt.

Neue Krebstherapie mobilisiert die Immunabwehr
Boston -  Eine kombinierte Immuntherapie gegen Krebs hat in ersten klinischen Versuchen Erfolge gezeigt, berichtet das Fachmagazin "PNAS". Die Therapie beruht auf einer am Dana Faber Cancer Institute in Boston entwickelten Impfung, bei der Patienten mit Zellen ihres eigen Tumors geimpft werden. Die Tumorzellen werden dem Körper entnommen und vor der Rückgabe so verändert, dass sie das Molekül CM-CSF produzieren. Dieses stimuliert das Immunsystem.


Unterstützt wurde die Impfung mit einer Gabe von Antikörpern gegen eine Substanz, die die Aktivität von Immunzellen bremst: Diese Bremse ist das Molekül CTLA-4. T-Zellen tragen es auf ihrer Oberfläche, sobald sie aktiviert wurden. Es ist quasi ein Ausschalter und verhindert, dass die Immunantwort länger als nötig andauert und so körpereigene Zellen angreift. Bei einer Impfung gegen den eigenen Tumor ist jedoch eine länger andauernde Immunreaktion gewünscht. Dass dies durch Antikörper gegen das Oberflächenmolekül CTLA-4 erreicht werden kann, hatten Forscher der University of California in Berkeley im Tierversuch bereits gezeigt.


Für die nun veröffentlichte klinische Studie haben die Forscher aus Boston und Berkeley ihre Ideen gebündelt und die beiden Therapien hintereinander an fünf Patienten mit bösartigem Haut- beziehungsweise Eierstockkrebs erfolgreich getestet: Die Antikörper gegen CTLA-4 verstärkten die Krebszellen zerstörende Wirkung der Impfung. Das Positive an dieser Therapie sei, so James Allison von der University of California, dass das Immunsystem dabei nur so lange stimuliert wird, bis die Antikörper abgebaut sind. Danach funktioniere das Immunsystem wieder normal, und der Patient behalte nur eine größere Zahl an Antitumor-T-Zellen zurück.
